摘要: 目的:探讨玄参提取物(SnPs)对局灶性缺血后脑组织的保护作用及对脑血流量的影响。方法:雄性 SD大鼠(242±s7)g 40只随机分5组(均n=8):假手术组、模型对照组、3个不同剂量 SnPs治疗组(1,5,10 mg.kg-1尾静脉注射)。大脑中动 脉线栓法建立局灶性脑缺血模型(MCAO),缺血24h后,用神经功能评分和梗死体积改变来评价 SnPs对脑缺血约治疗作用。另将 MCAO)模型缺血SD大鼠分成4组(模型对照组n=8,3个不同剂量1,5,10 mg.kg-1 SnPs治疗组均n=6)用于测脑血流 量,用激光多普勒仪分别在6个时间点(缺血前,缺血即刻,缺血后30 min和2 h,以及缺血2h再灌30min、2 h)记录双侧的皮层血流量。结果:脑缺血24h后。各SnPs 治疗组与模型对照组相比,脑梗死体积明显减少(P<0.05),神经功能均明显改善(P<0.01);但在此剂量范围内无明显的剂量依赖性。各SnPs 治疗组与模型组相比,缺血2 h后皮层CBF明显改善(P<005),其中5 mg.kg-1 剂量组对于缺血各时间点的血流改善均有显著作用(P<0.05)。结论:SnPs 对于脑缺血损伤具有保护作用,此作用 可能与提高脑血流量有关。
英文摘要: AIM: To investigate the protective eff.of of Scrophularia ningpoensis extracts ( SnPs ) on focal cerebral ischemic and the influcncc on cere- bral blood flow ( CBF ). METHODS: Male Sprague-Dawley rats were ubjected to middle cere-bral artery occlusion (MCAO) . Forty rats were ran-domly divided into five groups ( n = 8) , sham opcra-tion group, model group and SnPs-treatment groups ( l , 5, lO mg'kg-1) , respcctivcly. Neurobehavioral scres were determined 24 h after MCAO (grading of O to 4) . Ischemic lesion wolume was mcasured by 2.3 . 5 -triphenyltetrazolium chlo.ide t TTC) s,aining. CBF was monitored at six time points: before MCAO; instant. 0.5'h. 2 h aftcr MCAO and n.5 b, 2 h after repedusion performed (2 h afte. MCAO) in three SnPs-treated groups (l . 5, 1O mg'kg-1) and control group (received saline) . RESULTS: The SnPs-treated groups showed significant improvemcnt in neurological deficits compared with control group (P<0.01). Lesion volume was (15±s 4) % , (14±5) %, (15±3) % and (22±4) % ,in three treat- ed groups and control groups. respcctively. The le- sion volume was significantly decreased 24 h afte. MCAO in SnPs groups compared with the control gmup (P<:0.05) . In cnmparison to CBF decrease in control group, CBF was significantly improved in the dose of 5 mg'kg-1 at all points of time ( <0.05). in other two treated groups, CBF was obviously im- proved only at the time point of 2 h after MCAO (P<0.05).
